I just moved my wife's PC to another room one floor up in our house. Powerline extender is showing connectivity, but no internet connection from the router. Any suggestions?

is your outlet in the new room where are you placed your wife's PC on the same electrical circuit as the adapter's outlet? They all have to be on the electrical circuit or the TP-link will not work.
