I've just purchased a new home, over 3,000 sq. ft. solid brick construction.
Qu1) Does the first node of a mesh network have to stay connected by ethernet cable, or is it only during setup?
Qu2) I currently have 2 Wifi network, each with it's own Wifi unit, Can I have effectivly two sets of mesh nodes, one for each network?

The first main Deco always needs to be connected via an Ethernet cable.
Qu2) I currently have 2 Wifi network, each with it's own Wifi unit, Can I have effectivly two sets of mesh nodes, one for each network?
Do you want to create two Mesh networks via Deco units and each Mesh system has its own WiFi SSIDs?
If yes, you can refer to How to create a new network in the Deco app
