I bought this tp-link ac1300 mac archer t3u because my old iMac keeps dropping off the (uverse) wifi (hub is in basement, have repeater in the next room).

 

Once I activate this adaptor via its own menu, should I "Turn Wi-Fi Off" at the original menu?

By orginal menu i assume you are talking about the TP-Link adpater ultilty vs the built in adapter utlity.  The only thing yuo should have to do is remove any saved networks to the adapter so its not trying to connect to anything.  However I would say if you are not going to use the built in adapter utlity to just disable to be safe
